چکیده

This fall marks 20 years since the cloning of unc-119 was reported. Despite having a strong phenotype that makes animals somewhat difficult to grow and handle, unc-119 mutant rescue has become one of the most frequentlyused markers for C. elegans transformation. In this Commentary, I describe the history of how unc-119 rescue traveled through the worm community, contributing to the development of transgene methods in C. elegans.

Imprinting capacity of gamete lineages in Caenorhabditis elegans.

We have observed a gamete-of-origin imprinting effect in C. elegans using a set of GFP reporter transgenes. From a single progenitor line carrying an extrachromosomal unc-54::gfp transgene array, we generated three independent autosomal integrations of the unc-54::gfp transgene. The progenitor line, two of its three integrated derivatives, and a nonrelated unc-119:gfp transgene exhibit an impri...

UNC-119 suppresses axon branching

Typically, mature neurons are polarized cells composed of a dendritic arbor and a single axon (reviewed by Craig and Banker, 1994; Higgins et al., 1997). This polarity becomes apparent early during development when the neuron extends a growth cone to form a single unbranched axon (reviewed by Goodman, 1996; Goodman and Tessier-Lavigne, 1997; Mueller, 1999). In some neuronal populations collater...

GTP-binding of ARL-3 is activated by ARL-13 as a GEF and stabilized by UNC-119

Primary cilia are sensory organelles indispensable for organogenesis and tissue pattern formation. Ciliopathy small GTPase ARLs are proposed as prominent ciliary switches, which when disrupted result in dysfunctional cilia, yet how ARLs are activated remain elusive. Here, we discover a novel small GTPase functional module, which contains ARL-3, ARL-13, and UNC-119, localizes near the poorly und...

Identification and cloning of unc-119, a gene expressed in the Caenorhabditis elegans nervous system.

A spontaneous mutation affecting locomotion of the nematode Caenorhabditis elegans has been mapped to a new gene, unc-119. Phenotypic characterization of the mutants suggests the defect does not lie in the musculature and that the animals also have defects in feeding behavior and chemosensation. unc-119 has been physically mapped relative to a previously identified chromosomal break in linkage ...
